生物学的記述

Specificity Acetyl-Histone H2B (Lys15) Antibody (Rabbit mAb) [M20B13] detects endogenous levels of total Histone H2B protein only when it is Acetylated at Lys15.
Background Acetylation of histone H2B at lysine 15 (H2BK15ac) is a modification of the H2B N-terminal tail. It works in a reciprocal switch with phosphorylation of the neighboring serine 14 (H2BS14ph), and this switch separates the chromatin of living cells from that of apoptotic cells. H2BK15ac is common in the nuclei of non-apoptotic cells and becomes scarce in apoptotic nuclei. In parallel, H2B phosphorylated at Ser14 builds up and associates with soluble, fragmented DNA in apoptotic nuclei. This loss of K15 acetylation and gain of S14 phosphorylation occurs in rat thymocytes driven into apoptosis by glucocorticoids, both in culture and in animals treated with glucocorticoids. It also occurs in a rat hepatoma cell line induced to die by UV-C irradiation or Fas ligand. Deacetylation of K15 is required before S14 can be phosphorylated, so removing the acetyl group from K15 is a prerequisite for the apoptotic phosphorylation of the neighboring residue. Blocking histone deacetylases during glucocorticoid treatment preserves this acetylation state and suppresses both S14 phosphorylation and DNA cleavage between nucleosomes, while apoptosis itself still goes ahead. The kinase responsible for the apoptotic S14 mark is Mst1 (mammalian sterile twenty kinase). Caspase-3 cleaves Mst1, and this cleavage switches Mst1 on as a kinase that phosphorylates H2B at Ser14 both in vitro and in vivo. Ser14 phosphorylation appears only after caspase-3 has cleaved Mst1, and it marks apoptotic chromatin in species from frogs to humans. Together, deacetylation of K15 and caspase-activated, Mst1-mediated phosphorylation of S14 form an apoptotic modification switch on the H2B tail that is tied to DNA degradation during programmed cell death.
