生物学的記述

Specificity ALKBH3 Antibody (Rabbit mAb) [B14A24] detects endogenous levels of total ALKBH3 protein.
Background ALKBH3, also called hABH3, is a human homologue of the Escherichia coli AlkB dioxygenase. AlkB repairs alkylation damage in DNA by oxidative demethylation of 1-methyladenine and 3-methylcytosine, and demethylation by the AlkB dioxygenases is an important mechanism for repair of N-alkylated nucleotides. ALKBH3 and the related enzyme hABH2 are oxidative DNA demethylases. ALKBH3 also repairs RNA, which hABH2 does not do. The two human enzymes have different catalytic properties and different subnuclear localization patterns, so they play distinct roles in the cell’s response to alkylation damage. Little is known about how cells handle alkylation damage in RNA, and repair of RNA may be an important defense mechanism. In human cells ALKBH3 forms a complex with the activating signal cointegrator complex (ASCC). ASCC3, the largest subunit of ASCC, is a 3'-5' DNA helicase. Its activity is crucial for generating the single-stranded DNA on which ALKBH3 preferentially dealkylates. ALKBH3 and ASCC therefore work together to repair 3-methylcytosine in DNA. ALKBH3 is overexpressed in various cancers, and both ALKBH3 and ASCC are important for resistance to alkylation damage in tumor cell lines. In cell lines that depend on ALKBH3 and ASCC3, loss of either protein raises the level of 3-methylcytosine and reduces cell proliferation, with formation of pH2A.X and 53BP1 foci. These results provide a molecular mechanism by which ALKBH3 and ASCC maintain genomic integrity in a cell-type specific manner. Repair of DNA damage is essential for genome integrity, and repair deficiencies in mammals are associated with cancer, neurological disease and developmental defects.
