Specificity α-adaptin A Antibody [D21J17] detects endogenous levels of total α-adaptin A protein.
Background α-Adaptin is a crucial subunit of the adaptor protein complex 2 (AP-2), which belongs to the family of adaptor protein complexes involved in intracellular vesicle transport. α-adaptin forms part of the heterotetrameric AP-2 complex, which consists of two large subunits (α and β2), a medium subunit (μ2), and a small subunit (σ2). The α-adaptin subunit contains a C-terminal appendage domain that functions as a scaffolding platform for endocytic accessory proteins. It also possesses regions that bind polyphosphoinositide-containing lipids, positioning AP-2 on the plasma membrane. AP-2 operates primarily in clathrin-dependent endocytosis by linking clathrin lattices to cargo proteins and membrane lipids and facilitating the formation of clathrin-coated vesicles (CCVs). The α and σ subunits contribute to recognizing specific endocytosis signal motifs, such as [ED]-X-X-X-L-[LI], which enables selective cargo capture. AP-2 also plays roles in recycling synaptic vesicle membranes, post-endocytic trafficking, and possibly ARF6-regulated non-clathrin pathways. Dysfunction or mutation in AP-2 components, such as α-adaptin, disrupts vesicle formation and endocytosis.
