製品説明 Baohuoside I（Icariside II）は、イカリソウ（Epimedium koreanum Nakai）から単離されたフラボノイドで、抗炎症作用と抗がん作用を示します。これは、ROS/MAPK経路を介して細胞毒性効果を発揮する可能性があります。
in vitro Baohuoside I accelerates apoptosis through the mitochondrial apoptotic pathway, involving the increment of BAX/Bcl-2 ratio, dissipation of mitochondrial membrane potential, transposition of cytochrome c, caspase 3 and caspase 9 activation, degradation of poly (ADP-ribose) polymerase and the over-production of reactive oxygen species (ROS). In vitro studies with three leukaemia and three solid tumour cell lines show that this compound has cytotoxic properties, with IC50 values of between 2.8 and 7.5 μg/ml.
in vivo Baohuoside I is both anti-inflammatory and immunosuppressive, but unlike the cytotoxic and anti-rejection drugs it suppresses only B-cell (antibody) and not T-cell responses. This compound may have a role in therapy of those autoimmune diseases.

実験の流れ Cells (1×106 cells/ml) are exposed to Baohuoside I (6.25, 12.5, and 25 μM) for 24 h, washed with PBS, and then harvested with trypsin EDTA, counted, fixed with 75% ethanol and stored at 4℃ prior to DNA content analyses. Samples are resuspended in 1.0 ml hypotonic propidium iodide (PI)-solution (50 mg/ml PI, 0.1% sodium citrate, 0.1% Triton X-100, 0.1 mg/ml ribonuclease A) and incubated in the dark for 1 h. PI-stained samples are analyzed with a flow cytometer.
