Specificity Cleaved PARP (Asp214) Antibody [C3D4] detects endogenous levels of the large fragment (89 kDa) of PARP protein only when cleaved at Asp214.
Background Cleaved PARP (Asp214) is the caspase-3-generated cleavage product of poly(ADP-ribose) polymerase 1 (PARP-1), a nuclear enzyme central to DNA repair. PARP-1 consists of an N-terminal DNA-binding domain with zinc fingers (residues 1–214), a central automodification domain, and a C-terminal catalytic domain. During apoptosis, caspase-3 cleaves PARP-1 specifically between Asp214 and Gly215, resulting in a 24 kDa DNA-binding fragment and an 89 kDa catalytic fragment. This cleavage inactivates PARP-1’s DNA repair capacity by disrupting DNA binding and poly(ADP-ribosyl)ation activity, thereby preventing NAD+ depletion, limiting necrosis, and promoting orderly apoptotic progression. The presence of cleaved PARP is widely used as a hallmark of apoptosis, signaling caspase activation and cellular disassembly.
