| in vitro | ISO-1 significantly inhibits the cytokine activity in vitro. It inhibits tumor necrosis factor release from macrophages isolated from LPStreated wild type mice but has no effect on cytokine release from MIF-deficient macrophages. |
| in vivo | ISO-1 significantly increases the survival rate in severe sepsis induced by endotoxin or cecal ligation and puncture (CLP). It could prevent airway hyperresponsiveness (AHR) in mouse ovalbumin (OVA)-challenge models. This compound attenuated ozone-induced cell recruitment, cytokine release and AHR in the mouse but did not affect measures of emphysema. It protects photoreceptors and reduces gliosis in experimental retinal detachment. |

Macrophages were collected 2 h after activation with LPS and washed one time in PBS (pH 7.4). Nuclear extract was isolated using NE-PER Nuclear and Cytoplasmic Extraction Reagents. |
