製品説明 Methyl Gallate (Methylgallate, Gallic acid methyl ester, Gallicin) is a plant polyphenol with antioxidant, anticancer, and anti-inflammatory activities.
in vitro Methyl gallate inhibits Treg cell-suppressive effects on effector CD4+ T cells and Treg migration toward tumor environment. The expression of Treg surface markers including CTLA-4, CCR4, CXCR4, and glucocorticoid-induced TNFR are significantly suppressed upon methyl gallate treatment[2].
in vivo In tumor-bearing hosts, methyl gallate treatment substantially reduces tumor growth and prolonged the survival rate. In contrast, nu/nu mice do not show decreased tumor progression in response to methyl gallate. In addition, in tumor-bearing Treg-depleted mice, tumor growth and the survival rates are not changed by methyl gallate treatment, strongly suggesting that the main therapeutic target of methyl gallate in tumor suppression is related to modulation of the CD4+CD25+ Treg cell functions. In the spleen of tumor-bearing mice, methyl gallate treatment induces a significant decrease in the CD4+CD25+Foxp3high Treg cell population. Especially, the number of tumor-infiltrating CD25+Foxp3high Treg cells is significantly lower in methyl gallate-treated mice. methyl gallate can be used to reverse immune suppression and as a potentially useful adjunct for enhancing the efficacy of immune-based cancer therapy[2].

実験の流れ HUVECs are grown in 96-well micro plates in a final volume of 100 μl culture medium containing 104 cells per well. After allowing the cultures to grow to confluence, chemical and H2O2 treatments are followed with certain intervals. To determine the cell viability, XTT assay kit is used. Two cases of oxidative stress are applied to pre-incubated HUVECs with chemical for 1 h: the treatment with 1 mM H2O2 for 1 h and the treatment with 0.1 mM H2O2 for 48 h. Concentrations of chemical are varied from 0 to 0.5 mM.
