Specificity Phospho-STAT1 (Ser727) Antibody [F23C16] detects endogenous levels of total STAT1 protein only when it is Phosphorylated at Ser727.
Background Phospho-STAT1 (Ser727) is the serine-phosphorylated form of STAT1 at residue Ser727, a modification that plays a pivotal role in regulating STAT1’s transcriptional activity within interferon (IFN) signaling. STAT1, a latent transcription factor, contains several domains: an N-terminal oligomerization domain, a coiled-coil domain, a DNA-binding domain, an SH2 domain for dimerization via Tyr701 phosphorylation, and a C-terminal transactivation domain (TAD) harboring the Ser727 phosphorylation motif (PMSP), which is targeted by MAPKs such as p38. Phosphorylation at Ser727 occurs downstream of IFN-α/γ, cellular stress, or LPS, dependent on prior Tyr701 activation, and enhances STAT1’s interaction with coactivators like CBP/p300. This modification is essential for maximal transcriptional activation of IFN-responsive genes: it augments promoter recruitment and chromatin binding, supporting antiviral responses, macrophage activation, and inflammation. Aberrant Ser727 phosphorylation is linked to tumorigenesis and impaired IFN signaling by disrupting normal STAT1 activity.
