Specificity Thymine Dimer Antibody [E15K19] detects endogenous levels of total Thymine Dimer protein.
Background Thymine dimer (cis-syn cyclobutane thymine dimer, CPD) is the most abundant UV-B-induced DNA photolesion formed exclusively between adjacent thymine bases on the same strand via [2+2] cycloaddition, classified as a mutagenic non-coding lesion within the cyclobutane pyrimidine dimer family. The cis-syn isomer features covalent C5–C6 bonds forming a rigid cyclobutane ring that saturates the 5,6-double bonds of both thymines, eliminating aromaticity, bending the DNA helix by ~30°, unwinding the TT step by ~40°, widening both grooves, pinching the phosphate backbone, weakening 5'-T:A pairing, and inducing anomalous tilt/twist angles localized primarily to the lesion site while preserving overall B-form stacking. Thymine dimers block replicative DNA/RNA polymerases, triggering nucleotide excision repair (NER) via XPC/Rad4 recognition of helical distortion or photolyase direct reversal; translesion synthesis by Pol η inserts AA opposite TT to minimize mutations, but unrepaired lesions cause C→T transitions at dipyrimidine sites characteristic of skin cancers, including melanoma, basal/squamous cell carcinoma. Negative supercoiling enhances dimer formation by facilitating base unstacking.
