Background ULK1 (Unc-51 Like Autophagy Activating Kinase 1) is a serine/threonine protein kinase belonging to the UNC-51/Atg1 family, mammalian homolog of C. elegans unc-51 and yeast Atg1, essential for autophagy initiation and neuronal axon guidance. ULK1 (1058 amino acids, ~140 kDa) comprises an N-terminal kinase domain (residues 1-280) with conserved catalytic triad, central proline/serine-rich (PS) domain (281-500) mediating protein interactions, and C-terminal domain (501-1058) with Atg13/FIP200-binding motifs and regulatory phosphorylation sites (Ser317/Ser555/Ser777 by AMPK; Ser757 by mTOR). ULK1 forms a stable complex with Atg13, FIP200, and Atg101 that localizes to ER tubulovesicular regions/Atg9 vesicles upon nutrient deprivation; AMPK phosphorylation activates ULK1 kinase activity, inducing autophosphorylation and Atg13/Atg101/FIP200 phosphorylation to nucleate phagophore assembly, while mTORC1 phosphorylation inhibits complex formation. ULK1 forms clusters of over 30 molecules to initiate autophagosome biogenesis and also has non-autophagic functions, such as mediating growth cone endocytosis for NGF trafficking (via SynGAP/syntenin) and regulating mitophagy. Dysregulation of ULK1 is implicated in neurodegeneration through axon defects, cancer, where suppressed autophagy aids tumor survival, and metabolic disorders.
