in vitro	XEN445 shows good in vitro inhibitory potency against EL, and high selectivity over LPL and HL.
in vivo	In vivo, XEN445 displays good ADME and PK properties. In mice, this compound (30 mg/kg, p.o.) raises plasma HDL-cholesterol concentration.

キナーゼアッセイ	Measurement of lipase activities
キナーゼアッセイ	A mixed micelle fluorogenic phosphatidylcholine/Triton X-100 assay is used to titrate inhibitors against human and mouse EL. EL is diluted in PBS (1.4 nM final concentration) containing 14% DMEM, plus 2% glycerol and 40 μL is added to each well of a clear bottom 96-well plate containing inhibitors or vehicle alone (5 μL in 10% DMSO). The enzyme-inhibitor mix is incubated at room temperature for 30 min prior to the addition of substrate (5 μL of 10 μM bis-BODIPY-FL C11-PC in 0.1% Triton X-100) and enzyme activity determined using a fluorescence plate reader at excitation and emission wavelengths of 490 and 520 nm, respectively.
