in vitro Xylometazoline is an α-adrenoceptor agonist commonly used as nasal decongestant, exhibits highest potency at α2B-adrenoceptor subtype with EC50 of 99 μM. Xylometazoline binds at adrenoceptor subtypeα1A, α1B, α1D, α2A, α2B, α2C with IC50 of 0.08, 0.56, 0.45, 0.98, 1.8, 0.22μM, repectively.

キナーゼアッセイ Measurement of intracellular Ca2+ responses
The measurement of intracellular Ca2+ responses is performed by using an aequorin-EGFP fusion protein according to the method of CRET. In brief, harvested cells are loaded with 5 μM coelenterazine h for 2.5 h at room temperature. Suspensions of cells in assay buffer containing NaCl 150 mM, CaCl2 1.8 mM, KCl 5.4 mM, Hepes 10 mM, D-glucose 20 mM (pH 7.4) are used for luminometric determination of intracellular Ca2+ transients in 96-well plates in a Berthold Centro LB 960 luminometer. In the case of recording Ca2+ responses induced by adrenaline and noradrenaline, 1 mM ascorbic acid is added. Light emission is recorded 5 s prior and 25–60 s upon autoinjection of the agonist dissolved in assay buffer at a sampling rate of 2 Hz.
