    Background
    COMMD1, also known as MURR1 and the copper toxicosis gene product, is a ubiquitously expressed 21 kDa copper-binding protein that plays versatile roles in cellular homeostasis and protein regulation. COMMD1 consists of an N-terminal α-helical VARiable Domain (VARD) spanning amino acids 1-121 and a C-terminal domain containing the COMM motif (amino acids 125-190), separated by a protease-sensitive site; this arrangement facilitates oligomerization into high molecular weight complexes both in solution and when associated with phospholipid membranes. COMMD1 localizes to the nucleus, cytoplasm, and endocytic vesicle membranes, where it interacts with multiple key proteins, including the copper transporter ATP7B (implicated in Wilson disease), the delta subunit of the epithelial sodium channel (ENaC), cystic fibrosis transmembrane conductance regulator (CFTR), superoxide dismutase 1 (SOD1), XIAP, HIF-1α, and Cullin-RING ubiquitin ligases. COMMD1 regulates protein folding, stability, ubiquitination, and proteolysis, notably promoting ubiquitination and degradation of the NF-κB RelA subunit, thereby inhibiting its nuclear translocation and activity; importantly, COMMD1 also restricts HIV replication in resting T cells. Its dysregulation is linked to neurodegenerative diseases, cancer progression, and disorders of copper metabolism.
