    Human Nuclear Antigen, commonly referred to as Proliferating Cell Nuclear Antigen (PCNA), is a conserved homotrimeric protein that functions as a DNA sliding clamp essential for DNA replication and repair. It forms a ring-shaped complex comprising three identical subunits, each containing two similar domains linked by an interdomain-bridging loop, which creates a central channel that encircles DNA and facilitates processive polymerase activity. The protein’s outer surface contains hydrophobic pockets that mediate interactions with various partner proteins through conserved PIP-box motifs, enabling its role as a critical coordinator in DNA synthesis, repair pathways, and cell cycle regulation. Post-translational modifications such as ubiquitination and SUMOylation dynamically regulate PCNA's interactions and functional switching, which is vital for maintaining genome stability.
