    Background
    NEDD8 (neural precursor cell expressed developmentally downregulated protein 8) is a small ubiquitin-like protein (UBL) of the ubiquitin family, sharing about 60% sequence identity with ubiquitin and possessing a conserved ubiquitin-fold that is crucial for its conjugation activity. The mature NEDD8 protein consists of 76 amino acids, forming a globular β-grasp fold with an α-helix and four β-strands, a flexible C-terminal tail ending in Gly76 (which forms an isopeptide bond with lysine residues on target proteins), and surface residues that confer selectivity for NEDD8-activating enzymes. In a process called neddylation, which closely resembles ubiquitination, NEDD8 is activated by the E1 APP-BP1/Uba3 heterodimer, transferred to the E2 enzyme Ubc12, and then ligated by E3 ligases, mainly to cullin subunits of cullin-RING E3 ubiquitin ligases (CRLs). Neddylation of cullins triggers conformational changes that enhance E2-ubiquitin recruitment and substrate ubiquitination, thus promoting CRL activity. This modification regulates various cellular functions, including cell cycle progression (by degrading p27 and cyclin E), DNA damage responses, signal transduction (such as NF-κB and Wnt pathways), and protein homeostasis, but does not directly tag proteins for proteasomal degradation. Dysregulation of neddylation contributes to cancer (by stabilizing pro-proliferative proteins), neurodegeneration (via altered neuronal survival proteins), and other disorders like fibrosis.
