    Background
    NF-YA, a critical subunit of the heterotrimeric transcription factor Nuclear Factor Y (NF-Y), belongs to the family of sequence-specific DNA-binding proteins that recognize the CCAAT box motif found in the promoter regions of numerous eukaryotic genes. NF-YA partners with the NF-YB and NF-YC subunits, which together mimic histone proteins through a conserved histone-fold domain “handshake” motif, enabling the complex to bind DNA with high specificity and affinity. NF-YA contains two alpha helices essential for inserting into the DNA minor groove to provide sequence-specific recognition, whereas NF-YB and NF-YC primarily interact with the DNA backbone to stabilize binding. NF-Y serves as a pioneer factor, promoting chromatin accessibility, facilitating recruitment of transcription machinery, and regulating gene expression critical for cell growth, differentiation, and response to environmental stimuli. NF-YA’s sequence-specific DNA recognition also influences the transcription start site selection and modulates developmental processes including flowering in plants via regulation of FT gene expression.
