    Background
    RhoGDI (Rho GDP-dissociation inhibitor 1, RhoGDIα/ARHGDIA) is a regulatory protein of the RhoGDI family that governs the cycling of Rho GTPases (such as RhoA, Rac1, and Cdc42) between their inactive, cytosolic GDP-bound and active, membrane-associated GTP-bound states. RhoGDI features a flexible N-terminal regulatory domain (residues 1–69) that adopts a helix-loop-helix motif (residues 34–57) upon GTPase binding to occlude switch I/II regions, and a C-terminal immunoglobulin-like β-sandwich domain (residues 70–204) with a conserved hydrophobic pocket (lined by Ile177, Val184) for binding the geranylgeranyl lipid anchor; Arg66 bridges the two domains, stabilizing the inhibitory conformation. RhoGDI controls Rho GTPase activity by sterically blocking GEF access (inhibiting GDP dissociation), extracting prenylated GDP-bound GTPases from membranes through lipid sequestration, and shuttling them in the cytosol or delivering them to specific membrane sites for spatiotemporal activation. Through this regulation, RhoGDI orchestrates actin cytoskeleton dynamics, lamellipodia and ruffle formation, cell migration, polarity, and adhesion, integrating into Rho/ROCK/mDia1 and Rac/PAK/WAVE signaling pathways responsive to growth factors and integrins. Dysregulation of RhoGDI is implicated in cancer metastasis via hyperactive Rho signaling in breast and prostate cancers and neurodegeneration, such as Alzheimer’s disease, through disrupted neuronal morphogenesis.
