Background
USP18 (Ubiquitin-Specific Protease 18, also known as UBP43) is a key deubiquitinating enzyme that specifically removes ISG15 from conjugated target proteins, thereby regulating the cellular ISGylation state essential for antiviral immunity and immune homeostasis. USP18 features the canonical USP fold with finger, palm, and thumb domains, including a zinc-coordinated finger domain for structural integrity and a catalytic triad mediating its isopeptidase activity. USP18 acts as a critical negative regulator of type I interferon signaling by binding to the IFNAR2 subunit of the interferon receptor and blocking JAK1 recruitment and STAT phosphorylation, thus preventing excessive interferon-driven inflammation and autoimmunity. USP18 expression is upregulated by interferons and inflammatory cues, serving as a feedback inhibitor that balances robust antiviral responses with immune regulation. Dysregulation of USP18 is implicated in autoimmune diseases, cancer progression, and altered therapeutic outcomes.
