Background
5T4 (TPBG, trophoblast glycoprotein) is a vertebrate-specific type I transmembrane glycoprotein encoded by TPBG on chromosome 6q14-15, classified as an oncofetal antigen with restricted adult expression but upregulated in carcinomas. It comprises a 310-aa extracellular domain with seven leucine-rich repeats (LRRs, 24 aa each flanked by LRR-N/C) forming a rigid β-sheet core for protein interactions, seven N-glycosylation sites conferring protease resistance, a 20-aa transmembrane helix (356-376), and a 44-aa cytoplasmic tail ending in a PDZ-binding motif (Ser-Asp-Val) interacting with TIP-2/GIPC. 5T4 inhibits canonical Wnt/β-catenin signaling by allosterically disrupting Frizzled-LRP5/6 via DKK-1, while activating non-canonical Wnt/PCP-JNK pathway promoting cytoskeletal reorganization, EMT, enhanced motility/invasion, and CXCR4-mediated chemotaxis/angiogenesis. 5T4 expression correlates with poor prognosis in breast, gastric, colorectal, and ovarian carcinomas; it serves as CAR-T/ADC/bispecific antibody target due to tumor selectivity.
