    Background
    Desmoglein 1 (DSG1) is a transmembrane glycoprotein cadherin of the desmoglein subfamily within desmosomes, predominantly expressed in the superficial granular and cornified layers of stratified epithelia, where it provides mechanical strength. DSG1 comprises a signal peptide, prodomain with RRQKR cleavage site yielding a 1000-amino acid mature protein, extracellular region with four cadherin domains (EC1-EC4) featuring Ca²⁺-binding motifs for homophilic adhesion, a transmembrane domain that drives lipid raft association via length and bulkiness, and a cytoplasmic tail with intracellular anchor (IA), proline-rich, and desmoglein-specific regions binding plakoglobin, plakophilin, and Erbin. DSG1 mediates calcium-dependent intercellular adhesion by trans-interacting EC1 domains clustered in lipid rafts, linking via plaque proteins to keratin filaments for tissue resilience under stress; its cytoplasmic tail suppresses EGFR/Erk signaling independent of adhesion, promoting keratinocyte differentiation, stratification, and barrier maturation. DSG1 TMD properties regulate desmosome assembly dynamics, plakoglobin recruitment, and cell surface stability. DSG1 autoantibodies cause pemphigus foliaceus acantholysis, mutations lead to striate palmoplantar keratoderma, and downregulation (e.g., by IL-13) impairs the esophageal barrier in eosinophilic esophagitis.
