    Background
    EGR1 (Early Growth Response 1) is a zinc finger transcription factor belonging to the EGR family of immediate-early genes, characterized by three Cys2-His2 zinc finger domains that enable specific binding to EGR response elements (EBS) in target gene promoters to regulate transcription. EGR1 features an N-terminal activation domain, a central repressive region, and C-terminal DNA-binding zinc finger motifs. Its promoter contains serum response elements (SREs) and CArG boxes, which facilitate rapid induction by stimuli such as growth factors via MAPK/ERK signaling. EGR1 acts as a key mediator linking extracellular signals to nuclear gene expression, driving processes such as cell proliferation, differentiation, apoptosis, and EMT through targets like cyclin D1, SNAIL, SLUG, PTEN, and p21, often via RAS-RAF-MEK-ERK-ELK1 cascades activated by ER stress or growth factors. EGR1 exhibits dual roles in cancer, acting as both an oncogene that promotes tumor proliferation, invasion, metastasis, and angiogenesis (e.g., via VEGFA) and as a suppressor that induces apoptosis. It is also dysregulated in cardiovascular conditions through inflammation and remodeling.
