Background
Interferon-gamma (IFN-γ) is a type II interferon cytokine that plays a pivotal role in bridging innate and adaptive immunity by activating immune cells such as macrophages and natural killer (NK) cells to enhance their cytotoxic activity. IFN-γ functions by binding to its heterodimeric receptor complex, consisting of IFNGR1 and IFNGR2 subunits, which promotes receptor dimerization and activates the intracellular kinases JAK1 and JAK2. This leads to phosphorylation and activation of STAT1 transcription factors, which translocate to the nucleus to induce the expression of interferon-stimulated genes (ISGs), including antiviral proteins like PKR. IFN-γ enhances antigen presentation by increasing MHC class I and II expression and promotes cell-mediated immunity through the activation of T lymphocytes. It also stimulates macrophages to produce reactive oxygen species, nitric oxide, and inflammatory cytokines, supporting its role in defense against intracellular pathogens such as Chlamydia and Toxoplasma gondii. Dysregulation or deficiency of IFN-γ or its receptor leads to increased susceptibility to infections. Additionally, IFN-γ is involved in inflammatory diseases, including atherosclerosis, and exhibits complex roles in tumor immunity.
