Background
p73 is a member of the p53 family of transcription factors, exhibiting significant structural and functional homology to p53, and is expressed in multiple isoforms generated by alternative splicing and promoter usage. These include the transcriptionally active TAp73 isoforms, which promote apoptosis and cell cycle arrest, and the dominant-negative ΔNp73 isoforms, which inhibit both p53 and TAp73 activity. p73 is composed of an intrinsically disordered N-terminal transactivation domain, a proline-rich region, a central DNA-binding domain highly similar to that of p53, and a tetramerization domain, with the addition of a unique C-terminal sterile alpha motif (SAM) domain involved in protein–protein interactions and transcriptional regulation. In response to DNA damage, p73 is phosphorylated at Tyr99 by c-Abl, promoting nuclear translocation and activation of pro-apoptotic target genes (such as p53AIP1), while acetylation at Lys321 by p300 further modulates its transcriptional activity.
