    Background
    Phospho-SHP2 (Tyr542) refers to the tyrosine-phosphorylated form at residue Tyr542 of SHP2, also known as PTPN11, a non-receptor protein tyrosine phosphatase (PTP) belonging to the classical PTP family. SHP2 comprises two N-terminal SH2 domains (N-SH2 and C-SH2), a catalytic PTP domain, and C-terminal tyrosine residues including Tyr542 and Tyr580; phosphorylation at Tyr542 enables intramolecular interaction with the N-SH2 domain, relieving autoinhibition and exposing the active site. Phosphorylated Tyr542 also binds Grb2, acting as a scaffold to recruit signaling complexes independent of phosphatase activity. Phospho-SHP2 (Tyr542) activates upon growth factor stimulation via kinases like EphA2, promoting sustained ERK activation in Ras/MAPK pathways and modulating PI3K/AKT signaling through Gab1 dephosphorylation. It coordinates cellular responses to growth factors, cytokines, and hormones, regulating proliferation, differentiation, migration, and survival. Dysregulated Phospho-SHP2 (Tyr542) contributes to cancers via enhanced ERK signaling and to LEOPARD syndrome through aberrant MAPK activation.
